Ketotifen - belongs to the group of cycloheptathiophenones and has a pronounced antihistamine effect. Does not apply to bronchodilator antiasthmatics. The mechanism of action is associated with inhibition of the release of histamine and other mediators from mast cells, the blocking of histamine H1 receptors and the inhibition of the phosphodiesterase enzyme, as a result of which the level of cAMP (cyclic adenosine monophosphate) in mast cells increases.

 

Suppresses the effects of PAF (platelet-activating factor). When used alone, it does not stop asthmatic attacks, but prevents their appearance and leads to a decrease in their duration and intensity, and in some cases they completely disappear. Favorably affects the sputum production.

Pharmacokinetics

 

Absorption is almost complete, bioavailability is about 50% (due to the presence of the "first pass" effect through the liver). Passes through the blood-brain barrier.Penetrates into breast milk. Metabolised in the liver. It is excreted by the kidneys in the form of metabolites (the main metabolite is ketotifen N-glucuronide pharmacologically inactive). Within 48 hours the kidneys display the bulk of the dose (1% in unchanged form and 60-70% in the form of metabolites).

Inside, with food, adults - 1 mg 2 times a day in the morning and in the evening. If necessary, increase the dose to 2 mg 2 times a day.

 

Children from 3 years and older - 1 mg 2 times a day.

 

Duration of treatment - at least 3 months. The abolition of therapy is carried out gradually, within 2-4 weeks.

In addition, there are too many side effects, from which I already began to suffer more than from my pollinosis.First of all, it is, of course, drowsiness - I once did not fall asleep at the wheel! Then the constant dryness in the mouth is also a very unpleasant sensation. I also noticed that during the reception of the drug I became nervous, irritable, very much I eat and accordingly I start to recover. In general, I decided next year to give up ketotifen, I'll ask the doctor to prescribe me another medicine.

Ketotifen is an obsolete domestic drug. Adverse events do exceed the therapeutic effect. I like Kestin, but one tablet is not enough for a day. The best drug is Claritin !!! but it's too expensive! winked
