Colme - a drug for the treatment of alcohol dependence. The mechanism of action is due to the blockade of aldehyde dehydrogenase involved in the metabolism of ethanol. The blockade of this enzyme leads to an increase in the concentration of the metabolite of ethanol - acetaldehyde, which causes negative sensations (including flushing to the face, nausea, tachycardia, dyspnea), which make it extremely unpleasant to drink alcohol after taking the drug and lead to a conditioned reflex aversion to taste and smell of alcoholic beverages.
The sensitization effect of Colem on alcohol manifests itself earlier (about 45-60 minutes) and lasts shorter (about 12 hours) than the action of disulfiram. In contrast to Disulfiram cyanamide (the active substance of the drug Colme) does not have an antihypertensive effect and has a lower intrinsic toxicity. In addition, cyanamide has a greater specificity of action: unlike disulfiram, it blocks only aldehyde dehydrogenase and does not affect other enzymes, particularly dopamine-beta hydroxylase.
